RTTryptone Glucose Extract Agar is a standard methods medium for the cultivation and enumeration of microorganisms in dairy and bottled water products. This medium was adopted in 1948 by the American Public Health Association (APHA) for use in testing milk and dairy products. It is also currently the specified medium for the heterotrophic plate count procedure for testing bottled water.||Dehydrated Appearance: |Homogenous, free flowing, light beige ||Prepared Appearance: |Trace to slightly hazy, light beige to medium amber||pH: 7.0 ± 0.2||Test Procedure using Pour Plate Method:|1. Follow the pour plate method as described in Standard Methods (Vanderzant, C., and D. F. Splittstoesser (eds.). Compendium of methods for the microbiological examination of foods, 3rd ed. American Public Health Association, Washington, D.C.)|2. Incubate the inoculated medium in a humid atmosphere at 32 ± 1°C for 47-49 hours incubation for dairy samples or at 35 ± 0.5°C for at least 72 hours for water samples.|3.
