Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation. ||Applications: |Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.||Recommended Dilutions: |Immunohistochemistry (Formalin/paraffin): Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6, for 10-20 minutes followed by cooling at RT for 20 minutes. |Optimal dilutions to be determined by the researcher.||Recommended Positive Control: |Tonsil||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
