Macerozyme R-10® can be used in combination with cellulase "Onozuka R-10®" (cat. no. 16419) (1, 2) or with cellulase "Onozuka RS®" (cat. no. 16420) for the isolation of plant cells and especially for the isolation of metabolically active protoplasts.

Activity:
Pectinase: approx. 0.5 U/mg; unit definition: 1 unit catalyses the release of 1 µMol of reducing groups from pectic acid per minute at 25 °C, pH 4.5, calculated as galacturonic acid. Reducing groups are determined with an alkaline copper reagent (2).
Hemicellulase: approx. 0.25 U/mg; unit definition: 1 unit corresponds to 1 µMol of reducing groups released from beechwood xylan per hour at 37 °C, pH 5.5, calculated as xylose.
Cellulase: approx. 0.1 U/mg; unit definition: 1 unit catalyses the release of 1 µMol glucose from sodium carboxymethylcellulose per minute at 40 °C, pH 4.5; glucose is determined with an alkaline copper reagent (3).
Optimum pH: 3.5 - 7.0
Optimum temperature: 40 - 50 °C.
