-20°CDNA ladder was prepared from six different plasmids containing pUC, lambda phage and yeast genome sequences. Each plasmid was individually digested by the appropriate restriction endonucleases. The digests were then mixed. The marker yields the following 14 discrete fragments (in base pairs): 10,000, 8000, 6000, 5000, 4000, 3500, 3000, 2500, 2000, 1500, 1000, 750, 500, 250.||Band Concentration:|The band at 6000, 3000bp serves as a reference. 1ug of DNA Ladder Mix contains 70ng (~14% of total) of the 6000, 3000bp fragment. 1ug of DNA Ladder Mix contains 60ng (~12% of total) of the 1000bp fragment.||6X Loading Dye Solution (L3350):|Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 0.03% bromophenol blue, 0.03% xylene cyanol FF, 60% glycerol, 60mM EDTA.||Quality Control:|Analysis of 0.5ug (1ul) of the DNA ladder on agarose gel by ethidium bromide staining generates 14 descrete bands pattern.||Recommendations for Use: |1. Vortex gently just prior to use. |2. Prepare the DNA marker before loading by combining: |1ul (0.5ug) of D3930-11: DNA marker |1ul of L3350, Loading Dye Buffer, 6X |4ul of ddH2O.|3. Heat for 5 min at 65°C and then cool on ice for 3 min (see Note).|4. Apply the prepared amount (6ul) of the DNA marker on a 5mm lane of agarose gel.|5. Following electrophoretic separation on gel, visualize the DNA bands by ethidium bromide staining.||Storage and Stability:|May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
