It comes in a convenient 96-well format, with all the reagents necessary for 100 fluorescent HDAC activity measurements. In addition, the kit includes purified HDAC2 and a potent HDAC inhibitor, Trichostatin A, for the positive and negative control. The HDAC Fluorescent Activity Assay is based on the unique fluorogenic substrate and Developer combination. This assay method eliminates dealing with the radioactivity, extraction, and chromatography aspects of traditional assays. Using this kit, only two simple steps on a microtiter plate are needed to analyze the HDAC activity level. First, the HDAC fluorometric substrate, comprised of an acetylated lysine side chain, is incubated with a sample containing HDAC activity (e.g. HeLa nuclear extract or purified HDAC enzymes). The deacetylation will sensitize the substrate, causing further treatment with the Lysine Developer to produce a fluorophore that can then be measured using a fluorescence reader. ||Components: |H5109-67A: HDAC2 Recombinant Human 2x1ug|H5109-67B: HDAC Fluorogenic Substrate 1x50ul |H5109-67C: HDAC Developer 2x6ml| (contains Trichostatin A) |H5109-67D: Trichostatin A 1x100ul|H5109-67E: HDAC Assay Buffer 1x10ml|H5109-67F: Microtiter plate 1x96 wells||Applications: |Suitable for studying enzyme kinetics and screening small molecular inhibitors for drug discovery and HTS applications. ||Storage and Stability:|Aliquot to avoid repeated freezing and thawing and store at recommended temperature.
