-20°CS100 belongs to the family of calcium binding proteins such as calmodulin and troponin C. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains. Antibody to S100 stains Schwannomas, ependymomas, astrogliomas, almost all benign and malignant melanomas and their metastases. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes. S100 is a nervous tissue specific soluble protein made in glial cells. Present in sweat glands chongrocytes and myoepithelial cells. May be found as cytoplasmic, membranous, or mixed throughout the areas of tumor. Usually cytoplasmic when present in normal tissues. The diagnosis of Histocytosis X is confirmed by S100 staining. Suitable for use in immunohistochemical staining of formalin-fixed, paraffin-embedded tissues. S100 protein is highly solouble and may be eluted from frozen tissue duing staining.||Applications:|Suitable for use in Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Immunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30min at RT. No special pretreatment is required for staining of formalin/ paraffin sections.||Optimal dilution determined by the researcher.||Molecular Weight of Antigen:|~22kD (non-reduced)|~11kD (reduced)||Positive Control:|Melanoma or Schwannoma||Cellular Localization: Cytoplasmic||Storage and Stability: |May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add glycerol (40-50%) and freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.||Note: Applications are based on unconjugated antibody.
