4°C Do Not FreezeThe alpha 5 beta 1 integrin mediates cell adhesion and migration on fibronectin, a glycoprotein critical for normal vertebrate embryonic development. Indirect evidence reported to date suggests that this receptor also functions in the deposition of fibronectin matrices. Specificity of I7661-35Q was verified by its ability to immunoprecipitate a5b1 heterodimers from 125-I-surface labeled cells, reciprocal depletion of VLA-5 antigen from cell lysate with antiserum against the cytoplasmic domain of a5 integrin subunit, and immunoprecipitation of VLA-5 integrin from cells known to express this integrin. Cell lines tested to express a5b1, include: P19 cells, mouse melanoma B16F1 cells, mouse squamous carcoma SCC cells and thymocytes.| |Applications: |Suitable for use in Flow Cytometry, Western Blot, Immunoprecipitation and Inhibition of cell adhesion to fibronectin. Other applications not tested.||Recommended Dilutions:|Flow Cytometry: 10ug/ml|Western Blot: Non-denaturing gel|Inhibition of cell adhesion to fibronectin: 10ug/ml|Immunoprecipitation: 2ug from lysate of 10e6 cell equivalent. Note: That where cell adhesion to fibronectin involving other fibronectin receptors, such as VLA-4 (a4b1) integrin, is involved, inhibition of cell adhesion by this antibody alone may not be complete.|Optimal dilutions to be determined by the researcher.||Storage and Stability:|May be stored at 4°C. For long-term storage, aliquot and store at 4°C. Do not freeze. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
