Assay Principle:

The DetectX® Estriol ELISA Kit quantitatively measures Estriol in saliva, urine, extracted serum, extracted plasma, fecal extracts, and tissue culture media. This competitive ELISA has a run time of 2.5 hours. Please read the complete kit insert for more information before performing this assay. Use our provided Estriol standard to generate a standard curve for the assay.

Protocol Summary:

  • Place standards or diluted samples into the transparent microtiter plate provided, coated with goat anti-rabbit IgG antibody.
  • Add Estriol peroxidase conjugate and Estriol rabbit polyclonal antibody to the wells to start the immunological reaction.
  • Incubate the mixture covered at room temperature with shaking for 2 hours. The immunological reaction inversely correlates with the Estriol concentration in the sample.
  • After incubation, wash away the excess conjugate and introduce the TMB substrate, which reacts with the bound conjugate to produce a measurable signal.
  • Use a plate reader to detect the signal at 450nm and calculate the Estriol concentration based on the standard curve.

Background:

Estriol is a key hormone in the estrogen family known for its unique role during pregnancy. Unlike estradiol and estrone, Estriol is considered a weak estrogen, mainly produced through the hydroxylation of estradiol and estrone in the liver. In non-pregnant women, its levels are almost undetectable. However, during pregnancy, Estriol levels rise significantly, making it an important marker for monitoring fetal health and development.

Medical professionals routinely measure Estriol levels as part of prenatal screening tests. Monitoring these levels helps in assessing the well-being of the fetus, with abnormally low levels potentially indicating chromosomal or congenital anomalies. Additionally, Estriol has therapeutic applications in treating menopausal symptoms in some regions.

Recent research suggests that Estriol might also play a protective role in various health conditions, potentially impacting immune diseases, as well as bone and lipid metabolism. This expands its significance beyond reproductive health, opening avenues for further investigation in neurology and endocrinology.

The DetectX® Estriol ELISA Kit is an essential tool for researchers and healthcare professionals, offering accurate and efficient measurement of Estriol levels in diverse biological samples. This kit facilitates vital research in reproductive health, prenatal care, and potentially in broader medical fields.
