-20°Cp33 cdk2 associates with cyclin A in human cells. Kinase activity associated with cyclin A-cdc2 is found only in G2-phase. Cdk2 also complexes with cyclins E, D1 and D3. Cyclin E-cdk2 kinase is active in the G1- and S-phases of the cell cycle. It is important (as does cyclin A-cdk2) for the progression from G1- to S-phase. The levels of cyclin A-cdk2 is maximal at the G1/S transition. Both cdk2 and cyclin A are found associated with DNA in the initiation complex during replication. Rb protein acts as substrate for cdk2-cyclin E and/or cdk2-cyclin A in vivo. Cdk2 is activated and specifically localized to the nucleus during late G1-, S- and G2-phase.||Applications:|Suitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications not tested.||Recommended Dilutions: |Immunoprecipitation: 4ug Immunoprecipitates cdk2 from 500ug of mouse 3T3 nuclear extract.|Western Blot: 0.5-2ug/ml. HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with C2572-07. Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system.|Optimal dilutions to be determined by the researcher.||Recommended Secondary Antibodies:|I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo|I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo||Positive Antigen Control (Included):|H1838: HeLa Nuclear Extract||Storage and Stability:|May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receiot. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
