The IHC detection systems based on the polymers method consist of a polymer of an active enzyme linked to a secondary antibody specific for a target primary antibody. This format makes it possible to significantly increase the amount of enzyme present at the site of localization and therefore a greater signal by converting a greater quantity of substrate into precipitate. Moreover, this system allows a one-step protocol instead of two with the ABC system (biotinylated antibody + avidin-enzyme complex).
Antigen detection with primary antibody of the same species as the test tissue yields high background when indirect detection method is used. This severely limits the use of screening human antibody on human tissues. Our kits are designed for staining human primary antibodies on human tissues without background staining. The NeoKlear Human HRP-Polymer Detection kit provides special blocking buffers, polymeric HRP-linked secondary antibody as well as human primer in a ready to use system. This technology requires an overnight pre-incubation with primary antibody that results in excellent sensitivity and high specificity. It is a biotin-free system, therefore, overcomes the non-specific staining caused by streptavidin/biotin system due to endogenous biotins.
The most commonly used alkaline phosphatase substrates are Fast Red and Permanent Red. These 2 substrates form water-insoluble red precipitates when hydrolysed by alkaline phosphatase (AP). It is for this reason that they are commonly called chromogens. The localization of the target proteins is then carried out by microscopic visualization of the precipitates within the tissue.
