-20°CMast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation. ||Applications: |Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.||Recommended Dilutions: |Immunohistochemistry (Formalin/paraffin): Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6, for 10-20 minutes followed by cooling at RT for 20 minutes. |Optimal dilutions to be determined by the researcher.||Recommended Positive Control: |Tonsil||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
